Muscle Biopsy
The muscle biopsy service offers a comprehensive analysis of pediatric muscle biopsy specimens. A typical examination consists of a panel of special histochemical stains (Modified Gomori Trichrome, PAS with/without diastase, Oil Red-O), enzyme histochemical stains (NADH, SDH, Cytochrome Oxidase (COX), and ATPase), and fiber typing by immunohistochemistry (myosin heavy chain - fast and slow). When needed, electron microscopy is done on site. Immunohistochemistry for muscular dystrophy workup includes stains for: Adhalin (alpha-sarcoglycan), Beta-dystroglycan (B-dystroglycan), Dystrophins 1, 2, and 3, Gamma-sarcoglycan, and Merosin (laminin alpha 2). An Immunofluorescence panel ( IgG, IgA, IgM, c1q, C3, C4, albumin, and fibrinogen) is also available. Biopsies for cases of possible metabolic disease should have part of the specimen snap frozen in liquid nitrogen and stored at -70 degrees C. Such specimens may also be accompanied by a skin biopsy for ultrastructural analysis and production of a fibroblast culture.
